The cytotoxic and apoptotic effects of endemic Centaurea nerimaniae plant extract on HeLa and MDA-MB-231 cells

The aim of this study is to determine the effective concentration of Centaurea nerimaniae (C.n) plant methanol extract and to analyze its cytotoxic and apoptotic effects on HeLa and MDA-MB-231 cells for the first time. The cytotoxic effects of C. nerimaniae plant extract on HeLa and MDA-MB-231 cells were analyzed by realtime cell analyzer. Using the effective doses, active caspase-3 by immunohistochemical analysis, and apoptotic indexes by TUNEL assay and DAPI staining were evaluated. The differences were analyzed statistically between control and treatment groups. The cytotoxic effective doses of extract on HeLa and MDA-MB-231 cells were determined as 1,4 mg/ml and 3,6 mg/ml, respectively. We showed that the apoptotic cell indexes were significantly increased in treatment groups (p<0.0001). we observed the TUNEL positivity takes place only at the periphery of the nucleus in some MDAMB-231 cells. We determined the Caspase-3 immunopositive cells were significantly increased in the treatment groups (p<0.0001, p<0.001). Also we observed that the presence of positivity in nuclear or nuclear and cytoplasmic caspase-3 immune positive cells in treatment group of MDA-MB-231 cell line and the active caspase-3 was settled only around the nucleus in some cells. The results show that the treatment doses of the C. nerimaniae plant extract induced apoptotic cell death in HeLa and MDA-MB-231 cells. We also considered that the nuclear active caspase-3 in metastatic MDA-MB-231 cells may be depend on the response in apoptotic pathways and it can play a role on cell cycle proteins or in DNA fragmentation proteins in the nucleus.
