A Comprehensive Method Validation of Caffeine as an Anthropogenic Marker in Water Samples by LC-MS/MS

Özet

Caffeine is one of the most widely used pharmaceutics and preferred as an anthropogenic marker to indicate only human activities in water samples. Therefore, developing a fast and reliable analytical method for caffeine in water is necessary and crucial. In this study, it was aimed to develop and comprehensively validate a method conducted with Liquid Chromatography- Tandem Mass Spectrometry system. Validation parameters such as selectivity, analytical sensitivity, limit of detection and quantification, linearity, precision, accuracy and robustness were studied beside uncertainty. Correlation coefficient of calibration curve was found 0.997 whilelimit of detection and limit of quantification levels were measured as 0.165 and 0.548 ng/mL respectively. Repeatability studies were found below 10% RSD. Accuracy was between 87-109% in different days and at different concentrations in the range of 0.5-100 ng/mL.After the method validation, tap water samples (n=6) were enriched with concentrations of 50 ng/mL in 50 mL and solid phase extraction (SPE) recovery was found as 93.3%. This is the first comprehensively developed and validated method published in our country to detect caffeine in water with high precision, accuracy and recovery. The method can be easily applied to sediment, ground or seawater samples besides surface water and wastewater.
