Dünya genelinde Caryophyllaceae familyasına ait 105 cins ve 3000 tür bulunurken Herniaria

Tourn. ex L. cinsi de bu familyaya dahil olup 52 tür içermektedir [1]. Geleneksel olarak

Herniaria türleri, safra diskinezisi ve ürolitiyazis tedavisinde kullanılırken bu türlerin başlıca

saponinler, flavonoitler ve kumarinler açısından zengin oldukları bildirilmiştir [2]. Literatür araştırmaları sonucunda Herniaria pisidica ve Herniaria saxatilis türleri üzerinde daha önce hiçbir aktivite çalışması yapılmadığı tespit edilmiştir. Bu kapsamda ülkemizde yetişen

Herniaria pisidica ve Herniaria saxatilis bitki örneklerinin %80 metanol ve su ekstreleri hazırlanarak YBSK/KS/KS ile fenolik bileşiklerinin analizi ile insan meme ve kolon kanseri hücre hatlarındaki sitotoksik etkilerinin incelenmesi amaçlanmıştır. Ekstrelerin MCF-7 ve

COLO-205 hücre hatlarındaki sitotoksik aktivitesi MTT yöntemi ile belirlenmiştir. MCF-7 hücre hattına karşı en aktif ekstre Herniaria pisidica infüzyon ve COLO-205 hücre hattına karşı en aktif ekstre Herniaria pisidica %80 metanol ekstresi olarak bulunmuştur. IC50 değerleri incelendiğinde MCF-7 hücre hattı, test edilen ekstrelerin sitotoksik etkilerine karşı genel olarak dirençli bir profil sergilerken bu hücre hattına karşı aktivite gösteren tek ekstre 374,162 ± 27,53 µg/mL IC50 değeri ile Herniaria pisidica infüzyon ekstresi olmuştur. COLO-205 hücre hattı üzerinde yapılan değerlendirmede ise en güçlü sitotoksik etkiyi 300,75 ± 31,45 µg/mL IC50 değeri ile Herniaria pisidica %80 metanol ekstresi göstermiştir. YBSK/KS/KS ile yapılan analiz sonucu kinik asit, klorojenik asit, rutin, izokersitrin, hesperidin, nikotiflorin tayin edilmiştir. Elde edilen sonuçlar, Herniaria pisidica ve Herniaria saxatilis türlerinin fenolik bileşikler bakımından zengin olduğunu ve sitotoksik aktivite açısından potansiyel doğal kaynaklar olabileceğini göstermektedir.

While there are 105 genera and 3,000 species belonging to the Caryophyllaceae family worldwide, the genus Herniaria Tourn. ex L. is also part of this family and comprises 52 species [1]. Traditionally, Herniaria species have been used in the treatment of biliary dyskinesia and urolithiasis, and these species have been reported to be rich in saponins, flavonoids, and coumarins [2]. A review of the literature revealed that no previous studies on the bioactivity of Herniaria pisidica Brummitt and Herniaria saxatilis Brummitt had been conducted. In this context, the aim of this study was to prepare 80% methanol and water extracts from Herniaria pisidica and Herniaria saxatilis plant samples grown in Türkiye, analyze their phenolic compounds using HPLC/MS/MS, and investigate their cytotoxic effects on human breast and colon cancer cell lines. The cytotoxic activity of the extracts on the MCF-7 and COLO-205 cell lines was determined using the MTT assay. The most active extract against the MCF-7 cell line was the Herniaria pisidica infusion, and the most active extract against the COLO-205 cell line was the Herniaria pisidica 80% methanol extract. Upon examination of the IC50 values, the MCF-7 cell line generally exhibited a resistant profile against the cytotoxic effects of the tested extracts; the only extract showing activity against this cell line was the Herniaria pisidica infusion extract, with an IC50 value of 374.162 ± 27.53 µg/mL. In the evaluation conducted on the COLO-205 cell line, the 80% methanol extract of Herniaria pisidica exhibited the strongest cytotoxic effect, with an IC50 value of 300.75 ± 31.45 µg/mL. Analysis using YBSK/KS/KS identified quinic acid, chlorogenic acid, rutin, isoquercitrin, hesperidin, and nicotiflorin. The results indicate that the species Herniaria pisidica and Herniaria saxatilis are rich in phenolic compounds and may serve as potential natural sources for cytotoxic activity.
