Effects of adding bull seminal plasma to ram semen extenders on post-thaw spermatozoa motility and morphology

Özet

Semen was obtained from five Kivircik rams by electro-ejaculator and the seminal plasma was obtained by centrifuging the semen of a Holstein bull, which was collected by an artificial vagina, at 3000 rpm for 10 minutes. Pooled ram semen was split into three group. After the addition of extenders the final volume rates of bull seminal plasma in groups 1st, 2nd and 3rd were 0%, 7.5% and 15% respectively. Extended samples were cooled to +5°C, and following glycerolisation (5% glycerol v/v) and equilibration the samples were frozen in 0.25 ml straws (5 straws fro each group) at 110°C. The straws were kept in liquid nitrogen at -196°C for a month. The straws were thawed in a 37°C water bath for 30 seconds. Motility and morphologic defect rates were evaluated at post-thaw. The highest post-thaw motility was 30.90±12.47% in the group 2 containing 7.5% bull seminal plasma. It was determined that acrosomal and total morphology were kept best in the seminal plasma added 2nd and 3rd groups with values 56.20±10.74% - 61.00±9.27% and 57.60±13.08% - 63.60±11.28%. The difference between the groups and the control group was significant statistically (P<0.05). It was concluded that bull seminal plasma had beneficial effects on the freezing and post-thaw motility and morphology of ram semen when added to semen extenders.
