Valproic acid (VPA) is a drug that is frequently used worldwide in the treatment of epilepsy. Depending on
long-term use, VPA causes various complications such as coagulopathies, VPA-induced hepatotoxicity and
encephalopathy. Moringa oleifera Lam. (Family: Moringaceae) has been showed that it has antitumor, antiinflammatory, antiulcer, antispasmodic, diuretic, antihypertensive, antidiabetic and hepatoprotective activities.
The current study was designed to investigate the effects of Moringa leaves extract (70% ethanol) on
antioxidant system against VPA-induced oxidative damage in tongue tissues of rats. In the study, female
Sprague Dawley rats were randomly divided into four groups. Group I: control group (n=8); Group II: animals
given only 70% ethanol extract of moringa leaves for 15 days (0.3 g/kg b.w. /day) (n=8); Group III: animals
that received only sodium valproate for 15 days (0.5 g/kg b.w. /day) (n=15); Group IV: animals administered
with similar dose of sodium valproate + moringa extract for 15 days (n=12). Moringa extract and sodium
valproate were administered orally. Group I were orally given a similar dose of physiologic saline. At the end
of the experiment, tongues tissues were taken. Total antioxidant (TAS), total oxidant (TOS), reactive oxygen
(ROS), oxidative stress index (OSI), glutathione (GSH), lipid peroxidation (LPO) levels, superoxide dismutase
(SOD) and glutathione peroxidase (GPx) activities were determined in tongues tissues. In VPA groups, TOS,
ROS, OSI, LPO levels were found to increase while TAS and GSH levels and SOD and GPx activities
decreased. Administration of Moringa oleifera extract reversed these changes. These results indicated that
Moringa oleifera leaf extract had a protective effect on tongue tissues damage of VPA.
