Özet

Malonic acid (MA), methylmalonic acid (MA) and ethylmalonic acid (EMA) are diagnostic metabolites for different inborn metabolic diseases. These acids accumulate in urine of patients with malonic aciduria, methylmalonic aciduria and ethylmalonic aciduria. This study proposes a rapid and simple capillary electrophoretic (CE) with contactless conductivity detection (CDD) method was developed for the separation and the simultaneous determination of these acids in urine samples. The Box-Behnken design (BBD) was successfully employed for experimental design and analysis of the results. The effect of buffer pH, buffer concentration and CTAB concentration on resolution was studied and optimized using response surface methodology. Under the optimum conditions three acids were detected in less than 3 min. The developed method was applied to the determination of MA, MMA and EMA in urine samples.
